Modified vaccinia virus Ankara (MVA) has been widely tested in clinical trials as recombinant vector vaccine against infectious diseases and cancers in humans and animals. However, one biosafety concern about the use of MVA vectored vaccine is the potential for MVA to recombine with naturally occurring orthopoxviruses in cells and hosts in which it multiplies poorly and, therefore, producing viruses with mosaic genomes with altered genetic and phenotypic properties. We previously conducted co-infection and superinfection experiments with MVA vectored influenza vaccine (MVA-HANP) and a feline Cowpox virus (CPXV-No-F1) in Vero cells (that were semi-permissive to MVA infection) and showed that recombination occurred in both co-infected and superinfected cells. In this study, we selected the putative recombinant viruses and performed genomic characterization of these viruses. Some putative recombinant viruses displayed plaque morphology distinct of that of the parental viruses. Our analysis demonstrated that they had mosaic genomes of different lengths. The recombinant viruses, with a genome more similar to MVA-HANP (&gt;50%), rescued deleted and/or fragmented genes in MVA and gained new host ranges genes. Our analysis also revealed that some MVA-HANP contained a partially deleted transgene expression cassette and one recombinant virus contained part of the transgene expression cassette similar to that incomplete MVA-HANP. The recombination in co-infected and superinfected Vero cells resulted in recombinant viruses with unpredictable biological and genetic properties as well as recovery of delete/fragmented genes in MVA and transfer of the transgene into replication competent CPXV. These results are relevant to hazard characterization and risk assessment of MVA vectored biologicals.
The Orthopoxvirus genus belongs to the Poxviridae family. The orthopoxviruses (OPXV) are viruses with large linear double stranded DNA genome (170 to 250kbp) (1). OPXV can infect vertebrates and insects (2). Among the OPXV that cause human diseases, Variola virus (VARV), vaccinia-like virus, Cowpox virus (CPXV) and Monkeypox virus (MPXV) are the most common (3–6). VARV is the causative agent of smallpox, a deadly viral disease, which was eradicated in 1980 as a result of a massive vaccination campaign (7). During the smallpox campaign, Vaccinia virus (VACV) was used as the smallpox vaccine and several VACV strains have been developed and used in different countries, such as New York City Board of Health (NYCBH) was used in the America, Tian tan in China, Ankara in Turkey, and Lister and modified vaccinia virus Ankara (MVA) in Europe (8–10).

This study examines the SENSE-TRI program’s effectiveness in improving the fundamental&#13;
literacy skills of grade 3 learners in insurgency-challenged Gombe and Adamawa states of&#13;
Nigeria during the COVID-19 lockdown. A quasi-experimental design was employed, with&#13;
400 participants equally divided between randomly selected schools and pupils from SENSE&#13;
intervention schools (the treatment group) and a counterfactual group from schools and&#13;
pupils not participating in the SENSE-TRI program (the comparison group). The learners’&#13;
performance in both groups was assessed by the abbreviated Early Grade Reading Assess ment (EGRA), focusing on tasks such as letter-sound identification, syllable sound identifi cation, familiar word reading, invented word reading, and reading comprehension. The TRI&#13;
program compared learners’ scores in the treatment group with those of learners in the&#13;
comparison group using Tobit regression models. The results revealed that socio demographic variables had no significant independent influence on the observed outcomes.&#13;
However, the scores showed a statistically significant improvement in the literacy abilities of&#13;
the treatment group on all parameters and tasks compared to the control group. This&#13;
improvement exceeded the initial SENSE baseline reading proficiency aggregate values,&#13;
indicating the program’s effectiveness in both challenging and normal circumstances as a&#13;
possible way out when no schools exist
L&#13;
iteracy, numeracy, and life skills are basic skills needed to&#13;
engage individuals and harness their inputs towards national&#13;
development, hence the varied responses from global and&#13;
national actors, especially in insurgency-ravaged areas such as&#13;
north-east Nigeria. The Nigerian northeast region has been&#13;
embroiled in the insurgency since 2002 when the Boko Haram&#13;
sect, which literarily translates that Western education is for bidden, took up arms against the state and education. The modus&#13;
operandi of driving their ideology has left in its wake massive&#13;
destructions of infrastructure and human displacements. Among&#13;
areas closed are learning spaces, which have been argued to&#13;
threaten the progress of children, youth, and adults, particularly&#13;
those from more disadvantaged families (Agarwala et al. 2022).&#13;
These identified actors are deeply committed to rebuilding the&#13;
northeast region plagued by insurgency and counter-insurgency&#13;
activities, which have led to school closures, destruction of social&#13;
infrastructure, massive displacements and disruptions, and the&#13;
like (Jacob and Ensign 2020).
